The selection and the precise choice of MiRNA


> Introduction

microarn1.jpgHuman central nervous system tumors are mostly gliomas (50%), comprising generally of two sub categories: glioblastomas and oligodendrogliomas. These tumors, which are the leading cause of morbidity and mortality of patients with glial tumours represent a significant public health problem, with a median survival of seven years for oligodendrogliomas, and a comparable figure of only one year for glioblastomas after treatment.There is thus a great need to develop a diagnostic method capable of reliably and consistently detecting and correctly diagnosing early glial tumors perfectly.

The only current means of diagnosis relies on biopsies; a process known to be inconsistent and open to errors in terms of interpretation (two different experts may offer different diagnoses on the same sample). This innovative technology has no current equivalent, and is a solution to this problem of diagnosis. It utilizes the identification and use of microRNA biomarkers (known to play a key role in breakdowns in cellular processes, including glial tumors ) to ensure a simple, sensitive, consistant and accurate diagnosis.


> Principles of the technology

The selection and the precise choice of these MiRNA have been carried out by molecular analysis based on blood and tissue samples, necessitating the following steps:

1- LMicroRNA (RNA non coding for around 22 bases) have been extracted from tumoral tissue and submitted to quality control checks
2- The microRNA are administrated by methods based on hybridisation (DNA chips) on PCR (RT-qPCR) or via sequencing (next generation sequencing)
3- Expression profiles of several microRNA are used according to tumor type
4- The analysis of tumor type is made by comparison to reference values (reference grid)

In the end, the miRNA most evident in the diagnosis are selected and held back for use in the diagnosis and specification of glial tumors
